GHRH analogues such as CJC-1295 and Tesamorelin mimic growth hormone releasing hormone and act on the pituitary GHRH receptor. Ghrelin-mimetic secretagogues such as Ipamorelin act on a different receptor entirely, the GHSR. Because the two pathways are distinct, they are frequently studied in combination — which is why the CJC-1295 and Ipamorelin blend exists as its own product.

The DAC variant of CJC-1295 carries a drug affinity complex that substantially extends its half-life relative to the non-DAC form. That is a meaningful pharmacokinetic difference and the two are not substitutes for one another in a study design. Ipamorelin is characterised in the literature as comparatively selective among the ghrelin mimetics, which is the usual reason it appears in research protocols over older secretagogues.
